Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.3590T>G (p.Ile1197Ser), citing Ambry Variant Classification Scheme 2023: The p.I1197S variant (also known as c.3590T>G), located in coding exon 29 of the TSC2 gene, results from a T to G substitution at nucleotide position 3590. The isoleucine at codon 1197 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
