NM_001018005.2(TPM1):c.36G>T (p.Lys12Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K12N variant (also known as c.36G>T), located in coding exon 1 of the TPM1 gene, results from a G to T substitution at nucleotide position 36. The lysine at codon 12 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
