Uncertain significance — the classification assigned by Ambry Genetics to NM_001127208.3(TET2):c.740C>G (p.Ser247Cys), citing Ambry Variant Classification Scheme 2023: The p.S247C variant (also known as c.740C>G), located in coding exon 1 of the TET2 gene, results from a C to G substitution at nucleotide position 740. The serine at codon 247 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
