Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_181486.4(TBX5):c.1030G>A (p.Glu344Lys), citing Ambry Variant Classification Scheme 2023: The p.E344K variant (also known as c.1030G>A), located in coding exon 8 of the TBX5 gene, results from a G to A substitution at nucleotide position 1030. The glutamic acid at codon 344 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
