NM_181486.4(TBX5):c.672A>T (p.Gln224His) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q224H variant (also known as c.672A>T), located in coding exon 6 of the TBX5 gene, results from an A to T substitution at nucleotide position 672. The glutamine at codon 224 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
