Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_152594.3(SPRED1):c.362A>G (p.Glu121Gly), citing Ambry Variant Classification Scheme 2023: The p.E121G variant (also known as c.362A>G), located in coding exon 3 of the SPRED1 gene, results from an A to G substitution at nucleotide position 362. The glutamic acid at codon 121 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
