Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_003114.5(SPAG1):c.1057_1059delinsGAAGAC (p.Lys353delinsGluAsp), citing Ambry Variant Classification Scheme 2023: The c.1057_1059delAAAinsGAAGAC variant, located in coding exon 9 of the SPAG1 gene, results from an in-frame deletion of AAA and insertion of GAAGAC at nucleotide positions 1057 to 1059. This results in the substitution of the lysine residue for two amino acids (ED) at codon 353. This amino acid position is not well conserved in available vertebrate species. In addition, this variant is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear.
