NM_005359.6(SMAD4):c.1654G>C (p.Asp552His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome; Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D552H variant (also known as c.1654G>C), located in coding exon 11 of the SMAD4 gene, results from a G to C substitution at nucleotide position 1654. The aspartic acid at codon 552 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
