Pathogenic for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_005902.4(SMAD3):c.30del (p.Ile11fs), citing Ambry Variant Classification Scheme 2023: The c.30delG (p.I11Sfs*2) alteration, located in exon 1 (coding exon 1) of the SMAD3 gene, consists of a deletion of one nucleotide at position 30, causing a translational frameshift with a predicted alternate stop codon after 2 amino acids. This variant is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on the available evidence, this alteration is classified as pathogenic.
