Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003002.4(SDHD):c.344T>G (p.Val115Gly), citing Ambry Variant Classification Scheme 2023: The p.V115G variant (also known as c.344T>G), located in coding exon 4 of the SDHD gene, results from a T to G substitution at nucleotide position 344. The valine at codon 115 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
