Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001283009.2(RTEL1):c.2534C>A (p.Ala845Glu), citing Ambry Variant Classification Scheme 2023: The p.A845E variant (also known as c.2534C>A), located in coding exon 26 of the RTEL1 gene, results from a C to A substitution at nucleotide position 2534. The alanine at codon 845 is replaced by glutamic acid, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
