Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001283009.2(RTEL1):c.2066G>A (p.Arg689Lys), citing Ambry Variant Classification Scheme 2023: The p.R689K variant (also known as c.2066G>A), located in coding exon 23 of the RTEL1 gene, results from a G to A substitution at nucleotide position 2066. The arginine at codon 689 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
