Uncertain significance — the classification assigned by Ambry Genetics to NM_017763.6(RNF43):c.2186T>G (p.Leu729Arg), citing Ambry Variant Classification Scheme 2023: The p.L729R variant (also known as c.2186T>G), located in coding exon 8 of the RNF43 gene, results from a T to G substitution at nucleotide position 2186. The leucine at codon 729 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
