NM_022474.4(PALS1):c.1012A>G (p.Ile338Val) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1012A>G (p.I338V) alteration is located in exon 8 (coding exon 6) of the MPP5 gene. This alteration results from a A to G substitution at nucleotide position 1012, causing the isoleucine (I) at amino acid position 338 to be replaced by a valine (V). Based on data from gnomAD, the G allele has an overall frequency of <0.001% (1/251312) total alleles studied. The highest observed frequency was 0.001% (1/113626) of European (non-Finnish) alleles. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
