Uncertain significance — the classification assigned by Ambry Genetics to NM_001166108.2(PALLD):c.1965-12570C>A, citing Ambry Variant Classification Scheme 2023: The p.A154E variant (also known as c.461C>A), located in coding exon 1 of the PALLD gene, results from a C to A substitution at nucleotide position 461. The alanine at codon 154 is replaced by glutamic acid, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
