Uncertain significance — the classification assigned by Ambry Genetics to NM_002577.4(PAK2):c.804C>G (p.Asp268Glu), citing Ambry Variant Classification Scheme 2023: The c.804C>G (p.D268E) alteration is located in exon 9 (coding exon 8) of the PAK2 gene. This alteration results from a C to G substitution at nucleotide position 804, causing the aspartic acid (D) at amino acid position 268 to be replaced by a glutamic acid (E). Based on data from gnomAD, the G allele has an overall frequency of 0.003% (1/31394) total alleles studied. The highest observed frequency was 0.007% (1/15424) of European (non-Finnish) alleles. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
