Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_053025.4(MYLK):c.2285G>A (p.Cys762Tyr), citing Ambry Variant Classification Scheme 2023: The p.C762Y variant (also known as c.2285G>A), located in coding exon 13 of the MYLK gene, results from a G to A substitution at nucleotide position 2285. The cysteine at codon 762 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
