NM_000249.4(MLH1):c.266A>T (p.Glu89Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E89V variant (also known as c.266A>T), located in coding exon 3 of the MLH1 gene, results from an A to T substitution at nucleotide position 266. The glutamic acid at codon 89 is replaced by valine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
