Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_014915.3(ANKRD26):c.4360T>G (p.Leu1454Val), citing Ambry Variant Classification Scheme 2023: The p.L1454V variant (also known as c.4360T>G), located in coding exon 30 of the ANKRD26 gene, results from a T to G substitution at nucleotide position 4360. The leucine at codon 1454 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
