Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.149A>G (p.Lys50Arg), citing Ambry Variant Classification Scheme 2023: The p.K50R variant (also known as c.149A>G), located in coding exon 2 of the NF1 gene, results from an A to G substitution at nucleotide position 149. The lysine at codon 50 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
