NM_194454.3(KRIT1):c.1243A>G (p.Ile415Val) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1243A>G (p.I415V) alteration is located in exon 13 (coding exon 9) of the KRIT1 gene. This alteration results from a A to G substitution at nucleotide position 1243, causing the isoleucine (I) at amino acid position 415 to be replaced by a valine (V). Based on data from gnomAD, the G allele has an overall frequency of <0.001% (1/251230) total alleles studied. The highest observed frequency was 0.003% (1/30612) of South Asian alleles. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
