Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_014727.3(KMT2B):c.43T>G (p.Ser15Ala), citing Ambry Variant Classification Scheme 2023: The c.43T>G (p.S15A) alteration is located in exon 1 (coding exon 1) of the KMT2B gene. This alteration results from a T to G substitution at nucleotide position 43, causing the serine (S) at amino acid position 15 to be replaced by an alanine (A). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is well conserved in available vertebrate species. This alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
