NM_001365951.3(KIF1B):c.3821A>G (p.His1274Arg) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H1228R variant (also known as c.3683A>G), located in coding exon 33 of the KIF1B gene, results from an A to G substitution at nucleotide position 3683. The histidine at codon 1228 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this variant is unclear.
