Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.1328G>T (p.Gly443Val), citing Ambry Variant Classification Scheme 2023. This variant lies in the MSH6 gene (transcript NM_000179.3) at coding-DNA position 1328, where G is replaced by T; at the protein level this means replaces glycine at residue 443 with valine — a missense variant. Submitter rationale: The p.G443V variant (also known as c.1328G>T), located in coding exon 4 of the MSH6 gene, results from a G to T substitution at nucleotide position 1328. The glycine at codon 443 is replaced by valine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. In addition, the CoDP in silico tool predicts this alteration will likely impair molecular function, with a score of 0.982 (Terui H et al. J. Biomed. Sci. 2013;20:25). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
