NM_005591.4(MRE11):c.1967T>C (p.Phe656Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F656S variant (also known as c.1967T>C), located in coding exon 17 of the MRE11A gene, results from a T to C substitution at nucleotide position 1967. The phenylalanine at codon 656 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
