Uncertain significance for Ataxia-telangiectasia-like disorder — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005591.4(MRE11):c.1967T>C (p.Phe656Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine with serine at codon 656 of the MRE11 protein (p.Phe656Ser). The phenylalanine residue is moderately conserved and there is a large physicochemical difference between phenylalanine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with MRE11-related conditions. ClinVar contains an entry for this variant (Variation ID: 485822). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
