NM_013372.7(GREM1):c.507A>T (p.Arg169Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R169S variant (also known as c.507A>T), located in coding exon 1 of the GREM1 gene, results from an A to T substitution at nucleotide position 507. The arginine at codon 169 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
