NM_000245.4(MET):c.174T>G (p.His58Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the MET gene (transcript NM_000245.4) at coding-DNA position 174, where T is replaced by G; at the protein level this means replaces histidine at residue 58 with glutamine — a missense variant. Submitter rationale: The p.H58Q variant (also known as c.174T>G), located in coding exon 1 of the MET gene, results from a T to G substitution at nucleotide position 174. The histidine at codon 58 is replaced by glutamine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6089 samples (12178 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.006% (greater than 20000 alleles tested) in our clinical cohort. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
