Uncertain significance for FGFR3-related chondrodysplasia — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000142.5(FGFR3):c.1850A>G (p.Asp617Gly), citing Genomenon Sequence Variant Interpretation Standards - Updated. This variant lies in the FGFR3 gene (transcript NM_000142.5) at coding-DNA position 1850, where A is replaced by G; at the protein level this means replaces aspartic acid at residue 617 with glycine — a missense variant. Submitter rationale: FGFR3 p.Asp617Gly (c.1850A>G) is a missense variant that changes the amino acid at codon 617 from Aspartic acid to Glycine. To our knowledge, this variant has not been reported in patients affected with an FGFR3-related disorder in the published literature. Functional studies have been reported (PMID:26992226). It is absent or not present at a significant frequency in gnomAD. In silico models predict that this variant is possibly or probably damaging. In conclusion, we classify FGFR3 p.Asp617Gly (c.1850A>G) as a variant of uncertain significance.
