Uncertain significance for FGFR3-related chondrodysplasia — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000142.5(FGFR3):c.1745G>T (p.Cys582Phe), citing Genomenon Sequence Variant Interpretation Standards - Updated. This variant lies in the FGFR3 gene (transcript NM_000142.5) at coding-DNA position 1745, where G is replaced by T; at the protein level this means replaces cysteine at residue 582 with phenylalanine — a missense variant. Submitter rationale: FGFR3 p.Cys582Phe (c.1745G>T) is a missense variant that changes the amino acid at codon 582 from Cysteine to Phenylalanine. To our knowledge, this variant has not been reported in patients affected with an FGFR3-related disorder in the published literature. Functional studies have been reported (PMID:26992226). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify FGFR3 p.Cys582Phe (c.1745G>T) as a variant of uncertain significance.
