Pathogenic for Myelodysplastic syndrome — the classification assigned by Yasuda Lab, Institute of Science Tokyo to NM_032638.5(GATA2):c.1051A>G (p.Asn351Asp), citing ACMG Guidelines, 2015. This variant lies in the GATA2 gene (transcript NM_032638.5) at coding-DNA position 1051, where A is replaced by G; at the protein level this means replaces asparagine at residue 351 with aspartic acid — a missense variant. Submitter rationale: Previously, a case of a GATA2 missense variant at the same position (c.1052A>G, p. N351S) developed MDS (Hofmann I et al. Biol Blood Marrow Transplant 2020;26:1124–30.). Thus, it was assumed that the c.1051A>G variant was causative in our case, being classified as ‘pathogenic’ according to the American College of Medical Genetics and Genomics/ Association for Molecular Pathology classification.
