Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000245.4(MET):c.1958C>T (p.Ser653Phe), citing Ambry Variant Classification Scheme 2023: The p.S653F variant (also known as c.1958C>T), located in coding exon 6 of the MET gene, results from a C to T substitution at nucleotide position 1958. The serine at codon 653 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
