While this deletion is not anticipated to result in nonsense mediated decay, it is expected to create a truncated protein product. Multiple missense variants located within the deleted region, have been determined to be pathogenic. This suggests that this region is critical for NF1 protein function and that the deletion of the NF1 protein may also be pathogenic. The patient's phenotype is specific for NF1. For these reasons, this variant has been classified as pathogenic.
