Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000245.4(MET):c.262C>G (p.Pro88Ala), citing Ambry Variant Classification Scheme 2023. This variant lies in the MET gene (transcript NM_000245.4) at coding-DNA position 262, where C is replaced by G; at the protein level this means replaces proline at residue 88 with alanine — a missense variant. Submitter rationale: The p.P88A variant (also known as c.262C>G), located in coding exon 1 of the MET gene, results from a C to G substitution at nucleotide position 262. The proline at codon 88 is replaced by alanine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6088 samples (12176 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.01% (greater than 10000 alleles tested) in our clinical cohort. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
