NM_000027.4(AGA):c.83C>T (p.Pro28Leu) was classified as Pathogenic for Aspartylglucosaminuria by Institute of Biochemistry, Faculty of Medicine, University of Giessen, citing ACMG Guidelines, 2015: P28 is a highly conserved amino acid located close to the N-terminus of the alpha subunit, being the 5th residue of the mature alpha subunit. Therefore, the P28L substitution could have an effect on the removal of the signal peptide, or it may affect the polypeptide conformation. The variant has been reported in a compound heterozygous AGU patient. Overexpression studies in HEK293T cells showed a reduced enzyme activity (20-30% of reference activity) and reduced processing of the precursor protein in subunits.
