This variant lies in the COL2A1 gene (transcript NM_001844.5) at coding-DNA position 1998, where A is replaced by C; at the protein level this means the protein sequence is unchanged (glycine at residue 666 retained) — a synonymous variant. Submitter rationale: A heterozygous de novo p.Gly666= synonymous variant was detected in exon 31 of the COL2A1 gene (NM_001844.5). This variant has never been observed in population databases (PM2). In silico algorithms (SpliceAI) predict that this variant does not have a splicing-disruptive effect (BP7). The variant was detected de novo in the patient (PM6). Based on this information, this variant is classified as a Variant of Uncertain Significance (VUS) according to ACMG criteria. The COL2A1 gene is associated with "Spondyloepiphyseal dysplasia, Stanescu type" in the OMIM database. It is thought that this syndrome can explain the skeletal dysplasia findings, including femoral head hypoplasia, observed in the patient.
