Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_144997.7(FLCN):c.379C>T (p.Arg127Trp), citing Ambry Variant Classification Scheme 2023: The p.R127W variant (also known as c.379C>T), located in coding exon 2 of the FLCN gene, results from a C to T substitution at nucleotide position 379. The arginine at codon 127 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
