This variant lies in the PTEN gene (transcript NM_000314.8) at coding-DNA position 809, where T is replaced by C; at the protein level this means replaces methionine at residue 270 with threonine — a missense variant. Submitter rationale: The variant is not observed in the gnomAD v4.1.0 dataset. Predicted Consequence/Location: Missense variant. In silico tool predictions suggest damaging effect of the variant on gene or gene product [REVEL: 0.96 (>=0.6, sensitivity 0.68 and specificity 0.92); 3Cnet: 0.89 (> 0.75, sensitivity 0.96 and precision 0.92)]. The same nucleotide change resulting in the same amino acid change has been previously reported to be associated with PTEN-related disorder (PMID: 27066848).A different missense change at the same codon (p.Met270Lys) has been reported to be associated with PTEN-related disorder (PMID: 31674007). However the evidence of pathogenicity is insufficient at this time. Therefore, this variant is classified as VUS according to the recommendation of ACMG/AMP guideline.
