Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004064.5(CDKN1B):c.7A>G (p.Asn3Asp), citing Ambry Variant Classification Scheme 2023: The p.N3D variant (also known as c.7A>G), located in coding exon 1 of the CDKN1B gene, results from an A to G substitution at nucleotide position 7. The asparagine at codon 3 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
