NM_004064.5(CDKN1B):c.386A>G (p.His129Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H129R variant (also known as c.386A>G), located in coding exon 1 of the CDKN1B gene, results from an A to G substitution at nucleotide position 386. The histidine at codon 129 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
