Although the exact breakpoints of this deletion are not known, it is predicted to remove the initiation codon and result in an absence of protein or a truncation of the encoded protein due to translation initiation at a downstream site. Loss-of-function variants in this gene are known to be pathogenic. A deletion, matching exon 1 of the SRD5A2 gene was found at a frequency of 1.7e-05 in 120780 control chromosomes in the gnomAD database (Structural Variants v4.1 dataset). Similar copy number variants have been observed in multiple individuals affected with 3-Oxo-5 alpha-steroid delta 4-dehydrogenase deficiency (e.g. Kocova_2019, Mao_2025). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
