Likely pathogenic for von Willebrand disorder — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000552.5(VWF):c.7548+1G>A, citing LabCorp Variant Classification Summary - May 2015: Variant summary: VWF c.7548+1G>A is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing and loss of VWF function. Several computational tools predict a significant impact on normal splicing: Two predict the variant abolishes a 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 8e-06 in 250946 control chromosomes. c.7548+1G>A has been observed in an individual affected with unspecified phenotype in a large cohort study for secondary ACMG and non-ACMG genetic findings (Saeidian_2024). These report(s) do not provide unequivocal conclusions about association of the variant with Von Willebrand Disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
