This variant lies in the BARD1 gene (transcript NM_000465.4) at coding-DNA position 344, where T is replaced by C; at the protein level this means replaces leucine at residue 115 with proline — a missense variant. Submitter rationale: The p.L115P variant (also known as c.344T>C), located in coding exon 3 of the BARD1 gene, results from a T to C substitution at nucleotide position 344. The leucine at codon 115 is replaced by proline, an amino acid with similar properties. This alteration was also detected on a 25-gene panel test in a woman who was diagnosed with breast cancer before age 50 (Tung N et al. Cancer. 2015 Jan;121:25-33). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
