This variant lies in the NBEA gene (transcript NM_001385012.1) at the canonical splice donor site of the intron immediately after coding-DNA position 7035, where T is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.7035+2T>C variant is not present in publicly available population databases like 1000 Genomes, EVS, gnomAD, Indian Exome Database or our internal database. This variant has neither been published in literature in individuals affected with NBEA-related conditions nor reported to the clinical databases like Human Genome Mutation Database (HGMD), ClinVar or OMIM, in any affected individuals. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant can disrupt the consensus splice site. In-silico pathogenicity prediction programs like HSF3.1, MutationTaster2, CADD, Varsome, Franklin etc predicted the variant to be likely deleterious, however these predictions were not confirmed by published functional/translational studies.
