Likely pathogenic for RNU2-2 related neurodevelopmental disorder — the classification assigned by Kasturba Medical College, Manipal, Kasturba Medical College, Manipal, Manipal Academy of Higher Education, Manipal, India to NR_199791.1:n.104T>C, citing ACMG Guidelines, 2015: A known variant, n.104T>C in RNU2-2 was observed in homozygous state in the proband (Leitão et al., 2026, Greene et al., 2026, ClinVar ID: VCV004532801.2). Sanger validation and segregation analysis showed that the variant is present in homozygous state in the proband and the similarly affected sibling and heterozygous state in the parents. This variant is present in three individuals in heterozygous state and absent in homozygous state in gnomAD population database (v4.1.0).
