Uncertain Significance for Li-Fraumeni syndrome — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000546.6(TP53):c.1160C>G (p.Thr387Arg), citing ACMG Guidelines, 2015. This variant lies in the TP53 gene (transcript NM_000546.6) at coding-DNA position 1160, where C is replaced by G; at the protein level this means replaces threonine at residue 387 with arginine — a missense variant. Submitter rationale: This missense variant replaces threonine with arginine at codon 387 of the TP53 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). Experimental studies have shown that this variant did not impact function in yeast transactivation assays (PMID: 12826609) or human cell growth suppression assays (PMID: 30224644). In a large international case-control study, this variant was reported in 2/60466 breast cancer cases and 1/53460 controls (PMID: 33471991). This variant has been identified in 1/251492 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
