Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003072.5(SMARCA4):c.4139G>A (p.Ser1380Asn), citing Ambry Variant Classification Scheme 2023: The p.S1380N variant (also known as c.4139G>A), located in coding exon 28 of the SMARCA4 gene, results from a G to A substitution at nucleotide position 4139. The serine at codon 1380 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
