Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001365276.2(TNXB):c.10114G>A (p.Gly3372Ser), citing LabCorp Variant Classification Summary - May 2015: Variant summary: TNXB c.10108G>A (p.Gly3370Ser) results in a non-conservative amino acid change located in the Fibronectin type-III repeat domain (IPR003961) of the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 1.4e-05 in 1605146 control chromosomes in the gnomAD database (v4.1 dataset). This frequency is not significantly higher than estimated for disease-causing variants in TNXB, allowing no conclusion about variant significance. To our knowledge, no occurrence of c.10108G>A in individuals affected with TNXB-related conditions and no experimental evidence demonstrating its impact on protein function have been reported.
