Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003072.5(SMARCA4):c.304G>T (p.Gly102Cys), citing Ambry Variant Classification Scheme 2023: The p.G102C variant (also known as c.304G>T), located in coding exon 2 of the SMARCA4 gene, results from a G to T substitution at nucleotide position 304. The glycine at codon 102 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
