Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001267550.2(TTN):c.81617T>C (p.Ile27206Thr), citing LabCorp Variant Classification Summary - May 2015: Variant summary: TTN c.73913T>C (p.Ile24638Thr) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 1.2e-05 in 245800 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.73913T>C has been observed in an individual affected with cardioskeletal clinical features observed in TTN-related conditions who harbored a large deletion of TTN in trans; however three other similarly affected family members (father and two brothers) harbored the large TTN deletion, but did not carry this missense variant (Gohlke_2024). Therefore, this report does not provide unequivocal conclusions about association of the variant with TTN-related conditions. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
