Submitter rationale: The p.K744E variant (also known as c.2230A>G), located in coding exon 14 of the SMARCA4 gene, results from an A to G substitution at nucleotide position 2230. The lysine at codon 744 is replaced by glutamic acid, an amino acid with similar properties. This variant has been detected in multiple individuals with no reported features of Coffin-Siris syndrome (Ambry internal data).
